Cautions

AST (and to a lesser extent ALP and ALT) may be significantly falsely elevated in the presence of hemolysis. Falsely low ALT results may occur in patients treated with sulfasalazine. Samples with glycine, monoethanolamine, and high concentrations of phosphate and chloride may interfere with ALP measurements.

Method Description

ALP

Photometric rate with p-nitro-phenylphosphate (pNPP) in the presence of 2-amino-2-methyl-1-propanol (AMP) at pH 10.4.

 

ALT

Photometric rate with alanine to α-oxoglutarate to form pyruvate and glutamate. This assay does not contain pyridoxyl-5-phosphate.

 

AST

Photometric rate with aspartate and α-oxoglutarate to form pyruvate and glutamate. This assay does not contain pyridoxyl-5-phosphate.
