Cautions

LD and AST (and to a lesser extent ALP and ALT) may be significantly falsely elevated in the presence of hemolysis. Falsely low ALT results may occur in patients treated with sulfasalazine.
Samples with glycine, monoethanolamine, and high concentrations of phosphate and chloride may interfere with ALP measurements. Total and direct bilirubin results are falsely decreased in the presence of hemolysis. In some cases of monoclonal gammopathies (especially Waldenstrom's macroglobulinemia with very high concentrations of IgM), total and direct bilirubin results may be unreliable. Total and direct bilirubin results may be falsely low in the presence of high concentrations of N-acetyl-p-benzoquinone imine (metabolite of Acetaminophen).

Method Description

ALP

Photometric rate with p-nitro-phenylphosphate (pNPP) in the presence of 2-amino-2-methyl-1-propanol (AMP) at pH 10.4.

 

ALT

Photometric rate with alanine to α-oxoglutarate to form pyruvate and glutamate. This assay does not contain pyridoxyl-5-phosphate.

 

AST

Photometric rate with aspartate and α-oxoglutarate to form pyruvate and glutamate. This assay does not contain pyridoxyl-5-phosphate.
